This position will be placed within the research project Cellular Neurochemistry Group led by Assistant Professor Wojciech Michno, part of the research group Molecular Geriatrics, Department of Public Health and Caring Sciences, at the Rudbeck Laboratory.

The research environment provides internationally recognized expertise in in vitro cell models, including transdifferentiation and organoid generation, as well as multiomics and spatial multiomics. The research is supported by funding from prestigious sources, including the Knut and Alice Wallenberg Foundation, Science for Life Laboratory (SciLifeLab).

Duties
The successful candidate will work in an interdisciplinary research environment focused on genetic engineering–based strategies for cell and gene therapy in neurodegenerative disorders. The work includes the design, execution, and analysis of experimental studies aimed at understanding and manipulating cell identity, function, and therapeutic potential.

The main duties include genetic engineering of mammalian cells using advanced CRISPR-based technologies, including CRISPR-Cas9, Cas12a, prime editing, and forward CRISPR screening approaches. You will establish, optimize, and apply protocols for pluripotent stem cell culture, reprogramming, and in vitro differentiation, including lineage reprogramming toward disease-relevant neural and organoid models.

The position also involves developing and applying molecular and cellular assays to evaluate genetic modifications, cell identity, and functional outcomes, using techniques such as qRT-PCR, targeted next-generation sequencing, flow cytometry, immunocytochemistry, and protein-based analyses. You will be responsible for experimental planning, data analysis, documentation, and interpretation of results.

In addition to laboratory research, the role includes contributing to the communication of scientific results through preparation of reports, figures, and manuscripts for publication in peer-reviewed journals, as well as presentations at internal meetings and scientific conferences. Collaboration with other researchers, including biologists, clinicians, and bioinformaticians, is an integral part of the position.

Requirements
This position requires a doctoral degree in biomedical engineering, molecular biology, biotechnology, chemical engineering, biomedicine, or a closely related field. A strong experimental background in molecular and cellular biology is essential.

You must have documented hands-on experience in genetic engineering of mammalian cells, particularly using CRISPR-based technologies (e.g. CRISPR-Cas9, Cas12a, prime editing, or CRISPR screening approaches). Proven experience with pluripotent stem cell culture (human iPSCs and/or ESCs) and cell differentiation or reprogramming is required.

The position requires solid practical knowledge of molecular biology techniques, including DNA/RNA/protein isolation, PCR/qRT-PCR, molecular cloning, immunocytochemistry, immunohistochemistry, Western blotting, ELISA, and viral or non-viral gene delivery methods (e.g. lentivirus, AAV, electroporation, chemical transfection).

Experience with mammalian cell culture is mandatory, and familiarity with flow cytometry (FACS) and sequencing-based readouts is expected. The candidate must be able to independently plan experiments, troubleshoot protocols, and manage multiple experimental workflows in parallel.

Good oral and written communication skills in English are required.

Personal characteristics are of great importance. You must be self-motivated, proactive, and structured, with a strong ability to work independently as well as collaboratively in multidisciplinary teams. You should demonstrate excellent analytical skills, attention to detail, and a high level of responsibility in laboratory work and documentation.

Additional qualifications

The following qualifications are considered meritorious:

  • Experience with lineage reprogramming, direct cell conversion, or cell-based therapeutic development
  • Experience with forward genetic screens, pooled CRISPR libraries, or high-throughput genetic perturbation methods
  • Experience with organoid models, particularly neural or cortical organoids
  • Experience with targeted or bulk NGS, including experimental design and data interpretation
  • Familiarity with cell-based therapies or translational research in neurodegenerative disorders
  • Experience with viral vector design, production, titration, and transduction (lentivirus and/or AAV)
  • Experience working in interdisciplinary or translational research environments

Strong pedagogical skills, the ability to mentor junior colleagues or students, and experience collaborating across scientific disciplines are also considered advantageous.
