The Department of Immunology, Genetics and Pathology at Uppsala University (www.igp.uu.se) has a broad research profile with strong research groups focused on cancer, autoimmune and genetic diseases. A fundamental idea at the department is to stimulate translational research and thereby closer interactions between medical research and health care. Research is presently conducted in the following areas: medical and clinical genetics, clinical immunology, pathology, neuro-oncology, vascular biology, radiation science and molecular tools. Department activities are also integrated with the units for Oncology, Clinical Genetics, Clinical Immunology, Clinical Pathology, and Hospital Physics at Akademiska sjukhuset, Uppsala. The department has teaching assignments in several education programmes, including Master Programmes, at the Faculty of Medicine, and in a number of educations at the Disciplinary Domain of Science and Technology. The department has a yearly turnover of around SEK 420 million, out of which more than half is made up of external funding. The staff amounts to approximately 345 employees, out of which 100 are PhD-students, and there are in total more than 700 affiliated people.

Project description: Responsible for projects regarding blood vessel function during embryogenesis and disease in mice. The project includes analysis in various genetically modified mouse models with advanced microscopy of the vascular tree in different stages of development, focused on but not limited to the vascular tree in the retina and CNS. Leica and Zeiss confocal microscopes are used for static and live imaging followed by image analysis using ImageJ, Volocity and Imaris, and statistical evaluation. Various treatments on mice are performed with subcutaneous, intravenous, intraperitoneal and intraocular administration of substances followed by perfusion and production of tissue, for example retina. Further analysis of cells in 2D or 3D culture, purified from mouse tissue or purchased, in some cases virus-infected. The cells are examined for gene and protein expression and ability to respond to stimuli in various cell biological assays by RNA sequencing and mass-spectrometry. The project ends with a data compilation for oral and written presentation.

Qualifications: Doctoral degree and postdoctoral research activity at a leading international level.
Documented extensive experience and skill in animal experimental work, genotyping, injections of various kinds, tissue fixation, sectioning, immunostaining with different reading methods with focus on the vasculature.
Advanced microscopy and image analysis.
Extensive experience and skill in genetic analysis methods and genetic tools for regulating gene siRNA, homologous recombination, Cre-loxp system).
Experience in constructing recombinant lentivirus.
Extensive experience and skill in biochemical and cell biological analysis methods (transcriptional analysis, immunoprecipitation, immunoblotting, immunostaining, analysis of different cell reactions, such as cell division, cell migration), 3D (explant) culture.
Extensive experience and skill in terms of oral and written presentation.
Ability to problem solve and develop the project independently.

Merits: Excellent credentials in the form of publications in top-ranked scientific journals. Extensive knowledge practically and theoretically about endothelial cells and blood vessel function.

Starting date: 01-01-2021 or as otherwise agreed.
