The Science for Life Laboratory (SciLifeLab) is collaboration between four universities in Stockholm and Uppsala: Stockholm University, Karolinska Institutet, the Royal Institute of Technology, and Uppsala University. The SciLifeLab is a national center for bioscience with a focus on health and environmental research. The center combines advanced technical know-how and state-of-the-art equipment with a broad knowledge in translational medicine and molecular biosciences. As part of this effort, the national platform for academic drug discovery and development was established within SciLifeLab – the SciLifeLab Drug Discovery & Development Platform (SciLifeLab DDD).

ADME of Therapeutics (ADMEoT) facility (Uppsala University Drug Optimization and Pharmaceutical Profiling-UDOPP) is organized at the Department of Pharmacy, Uppsala University and is part of the SciLifeLab DDD. The primary aim of ADMEoT (UDOPP) is to perform in vitro ADME (Absorption, Distribution, Metabolism and Excretion) studies and pharmaceutical profiling of hit collections and compound series during the lead generation/ optimization stage of the drug discovery process. The facility also provides preclinical characterization and performs bioanalytical services.

UDOPP (ADMEoT) is also affiliated to the Innovative Medicine Initiative (IMI), New Drugs for Bad Bugs (ND4BB) program, European Gram Negative Antibacterial Engine (ENABLE) consortium and to the IMI, ConcEPTION program. The Department of Pharmacy at Uppsala University is now recruiting a scientist with experience of bioanalysis and protein characterization to contribute to academic drug discovery projects within the SciLifeLab DDD. The position will be placed at SciLifeLab DDD, ADME of Therapeutics (UDOPP) facility at the Department of Pharmacy, Uppsala University.

The purpose of the position is to provide scientific expertise and contribute to the academic drug discovery (DD) projects ongoing at the SciLifeLab DDD. The major objective is to perform bioanalysis and characterization of biologics and therapeutic antibodies (mAb) using high resolution mass spectrometry (i.e. LC-IM-HR MS and Orbitrap MS) within drug discovery projects. The candidate’s responsibilities will include data acquisition, data analysis, generating written reports and presenting results at project meetings. The candidate’s role will also be to complement the work of the ADME team with analytical and biopharma expertise. Though the main focus will be on peptides and proteins, the candidate should be prepared for the analysis of small molecules and oligonucleotides, when the need arise.

Duties:

The selected candidate will be responsible for:

  • Sample preparation and quantitative analysis of therapeutic antibodies (mAb) from in vitro and in vivo experiments, using LC-MS/MS
  • Performing intact mass determination and peptide mapping analysis of proteins and antibodies
  • Performing post-translational modification (PTM) profiling‎ of therapeutic antibodies (mAb)
  • Developing, evaluating and implementing new technologies, with a focus on expanding the existing LC-IM-HR MS (liquid chromatography-ion mobility -high-resolution mass spectrometry) capacity
  • Sharing analytical expertise and protein knowledge with other members of the ADMEoT facility and SciLifeLab DDD
  • Generating written reports of the results to respective project forums in a timely and orderly manner
  • Contributing to the strategy for the analysis and characterization of therapeutic antibodies at the ADMEoT facility

Desired qualifications:

  • A PhD in bioscience with a specific focus on analysis and characterization of proteins using LC-MS/MS techniques
  • In-depth knowledge and several years of practical experience working with liquid chromatography coupled to mass spectrometry. Experience with the instruments currently in the lab, namely Aquity UPLC coupled to Synapt G2-Si and Dionex LC coupled to Q Exactive HF Orbitrap, is meritorious.
  • Experience with protein characterization and post-translational modifications
  • Detailed understanding of common proteomic assays and techniques, from basic proteolytic digestions to more complex quantitative and qualitative proteomic analysis
  • Hands-on experience of the analysis of test compounds in biological matrices (plasma and tissue homogenates), both in terms of sample preparation and the use of software for the data analysis (MassLynx, Unify, Xcalibur, Proteome Discoverer, Skyline)
  • Fluency in spoken and written English.
  • A documented academic track record and experience in biopharma are meritorious.
