Duties/Project description
It is known that in E. coli, initiation of chromosome replication starts bidirectionally from a single origin of replication (oriC) and is mediated by binding of several copies of the replication initiation protein DnaA together with the coordinating protein DiaA. The coordination of initiation with division cycle is generally described as being “complex”, and although a number of theoretical models have been suggested during the last 30 years, none of these are plausible in the light of the sensitivity seen in recent experiments.

To get to the mechanism, we will use large-scale screening to identify potential regulatory proteins and depending on the result we will be considering different quantitative models. The DuMPLING screen (Lawson et al. Mol. Syst. Biol. 2017) has been developed in the lab during the last couple of years and will be used in the initial phase. Potential candidates will then be investigated further using conventional KO/KD strategies. The results will be used to generate hypothesizes and new quantitative models for the mechanisms coordinating the replication and division cycle in E. coli.

Requirements
You have a passion for science and high ambitions to perform top quality research. You hold a master degree in a relevant subject area; you have good knowledge of single molecule imaging of fluorescent molecules in living cells and you are proficient in molecular cloning techniques, protein purification and florescence labelling. You have good programming skills. Personal qualities, ability to work independently and at the same time be able to interact with other researchers are very important. Excellent communication skills in written and spoken English are prerequisites.

Meriting
Experience with microfluidic techniques for studies of bacterial cells. Good knowledge of the bacterial cell cycle and regulation of replication initiation.
