The Department of Immunology, Genetics and Pathology at Uppsala University (www.igp.uu.se) has a broad research profile with strong research groups focused on cancer, autoimmune and genetic diseases. A fundamental idea at the department is to stimulate translational research and thereby closer interactions between medical research and health care. Research is presently conducted in the following areas: medical and clinical genetics, clinical immunology, pathology, neuro-oncology, vascular biology, radiation science and molecular tools. Department activities are also integrated with the units for Oncology, Clinical Genetics, Clinical Immunology, Clinical Pathology, and Hospital Physics at Akademiska sjukhuset, Uppsala. The department has teaching assignments in several education programmes, including Master Programmes, at the Faculty of Medicine, and in a number of educations at the Disciplinary Domain of Science and Technology. The department has a yearly turnover of around SEK 400 million, out of which more than half is made up of external funding. The staff amounts to approximately 340 employees, out of which 100 are PhD-students, and there are in total more than 600 affiliated people.

Duties:
The applicant will be responsible for a project concerning analysis of blood vessel leakage in healthy and tumor-bearing adult mice. A range of mouse strains will be analyzed. The project includes analysis with advanced microscopy, including two-photon and confocal microscopes, of the dermis vascular tree.
Various treatments are performed by subcutaneous micro-injection of substances into mice with fluorescent substances in the circulation, supplied by cannulation of the tail vein. Tumor cells are injected intradermally and allowed to grow into an established tumor. Microscopy is performed on the living tissue, with or without tumor, followed by image analysis (ImageJ, Volocity, Imaris or proprietary programs) and statistical evaluation. The fixed tissue can later be analyzed in the confocal microscope. It is also part of the project to take biopsies and determine the genotype in order to maintain the different recombinant mouse strains. Furthermore, analysis of endothelial cells in culture is included, purified from mouse tissue or purchased. The cells are examined for protein expression and ability to respond to stimuli in various cell biological assays. Isolated endothelial cells are also analyzed for transcript levels by single cell RNA sequencing. The project is concluded through data compilation for oral and written presentations.

Requirements:
Doctoral degree and postdoctoral research training at an international, high-quality level.
Considerable experience of and skill in animal experimentation, genotyping, injections of various kinds, dissection of organs (such as retina), tissue sectioning, and immunostaining.
Considerable experience and advanced skill in advanced microscopy using 2-photon and confocal microscopes, and image analysis and statistical analysis.
Considerable experience in biochemical and cell biology analysis methods (immunoprecipitation, immunoblotting, analysis of various cell reactions).
Great experience and skill in oral and written presentation in English.

Additional qualifications:
Excellent merits in terms of publications in leading scientific journals.
Considerable insights theoretically and practically concerning endothelial cells, blood vessel function in normal tissues and cancer.
