The Department of Cell and Molecular Biology is one of the most international, interdisciplinary and prominent biomolecular centers in Europe. The department consists of six research programs with approximately 130 employees. The advertised position is part of Prof. Suparna Sanyal's research group at the program of molecular biology (https://www.icm.uu.se/molecular-biology/sanyal-lab/)

Overall Description
The ribosome is the cellular protein factory, which translates genetic codes from messenger RNA (mRNA) into proteins. Protein synthesis also requires many translation factors, which play important roles in the process. In our lab, we try to understand how the ribosome and the translation factors work, how they perform the various steps of protein synthesis. This project aims to explore the mechanisms of protein synthesis in the cold bacteria that live between 0 - 15 ° C.

Work Description
As part of an active research group, you should conduct research to understand the molecular mechanisms of protein synthesis in cold bacteria Pseudoalteromonas haloplanktis TAC125. The assignment involves experimental design, execution, data collection and state-of-the-art analysis of the data. You will perform experiments independently but also interact with other members of the Sanyal group. Specific tasks include (but are not limited to)

  • Purification of ribosomes from Pseudoalteromonas haloplanktis TAC125
  • Cloning and purification of all translation factors from P. Haloplanktis TAC125
  • Fast kinetic analysis to study the mechanisms of different steps of translation
  • Biochemical data analysis
  • Documentation and dissemination of the knowledge in writing and oral forms

The doctoral student shall primarily devote himself/herself to his/her postgraduate studies full-time within the agreed research period. Other service at the department, which relates to education and administrative work, will not exceed 20%.

Requirements

  • Master's degree in molecular biology / molecular biotechnology / biochemistry or equivalent
  • Experience in molecular cloning, protein overexpression and purification
  • Knowledge of biochemistry (especially enzyme kinetics)
  • Basic knowledge of kinetic data analysis programs (e.g. Origin)
  • Excellent communication skills in English

For the appointment of the PhD student, the candidate must be accepted or admitted to postgraduate education.

Merits will be given to the candidate with

  • Experience in the production and purification of ribosomes.
  • Experience of using stopped-flow or quench flow
  • Experience of using HPLC
